Special Experimental Designs Using Laboratory Animals and Sample Size: Review. (English)

To achieve the scientific objectives in biomedical studies using laboratory animals, decision- making procedure of the appropriate experimental design and the sample size is important ethics, economic, and scientific. In this study, the special experimental designs such as Change-over, Cross-over, Switchback, Reversal and Latin square designs used in research involving laboratory animals have briefly introduced and their advantages are referred, and it is dwelled upon how the necessary minimum number of subject will be identified in the type of this experiments. The special experimental designs used in researches with laboratory animals have been briefly summarized on the simulated examples. In addition, when deciding the number of appropriate subjects, it is described to resource equation method, power analysis, and other practical approaches also taking into account factors such as data structure, the type of design and subject. In laboratory researches, it is emphasized that the special experimental designs produce scientifically more realistic estimates, and also minimize the needed number of subjects. Furthermore, when considering the restrictions imposed by the ethics problems, it is explained which the researches with laboratory animals are repeated in different the place, time, and conditions, also and the necessity of merging the results of the researches on the same topic by the meta-analysis. Laboratuvar hayvanlarının kullanıldığı biyomedikal çalışmalarda bilimsel amaçlara ulaşabilmek için uygun deney tasarımı ve örneklem hacmine karar verme işlemi etik, ekonomik ve bilimsel açıdan önem taşımaktadır. Bu çalışmada laboratuvar hayvanlarının yer aldığı araştırmalarda kullanılan Change-over, Cross-over, Switchback, Reversal ve Latin kare tasarımları gibi özel deney tasarımları kısaca tanıtılarak avantajlarından bahsedilmiş ve bu tip denemelerde yer alması gereken minimum denek sayısının nasıl belirlenebileceği konusu üzerinde durulmuştur. Laboratuvar hayvanları ile yapılan araştırmalarda kullanılan özel deney tasarımları simüle örnekler üzerinde kısaca tanıtılmıştır. Ayrıca uygun denek sayısına karar verirken veri yapısı, tasarım türü ve denek türü gibi faktörleri de dikkate alarak kaynak eşitlik yöntemi, güç analizi ve diğer pratik yaklaşımlar açıklanmıştır. Laboratuvar araştırmalarında özel deney tasarımlarının bilimsel açıdan daha gerçekçi tahminler ürettiği ve gerekli denek sayısını da en aza indirgediği vurgulanmıştır. Ayrıca etik sorunların getirdiği kısıtlamalar da dikkate alındığı zaman, laboratuvar hayvanları ile yapılan araştırmaların farklı yer, zaman ve koşullarda tekrar edilmesi ve aynı konu üzerinde yapılan araştırmaların sonuçlarının ise meta analizi ile birleştirilmesi gerekliliği açıklanmıştır.
